ssDNA for DNA tweezers assembly (Strand 2)
This part is initially transcribed into a non-coding RNA (ncRNA), and then it requires the chassis to possess a HIV reverse transcriptase to reverse transcribe the ncRNA into a single-strand DNA (ssDNA).
The ssDNA interacts with two other ssDNA produced by the parts BBa_K2618000 and BBa K2618002 to form a DNA tweezers. The DNA tweezers can be used to detect a potential biomarker (miRNA 25) of spinocerebellar ataxia type 3.
One end of this ssDNA consists of bases which are complementary to miRNA 25. Upon detection of miRNA 25 by the DNA tweezers, an intact G-quadruplex will be formed. When the G-quadruplex binds with hemin, it will exhibit a peroxidase activity, which can be detected by some colorimetric or fluorescent reagents like ABTS.
